| Finding Evidence of Infidelity Sexual activity often creates biological stains containing a mixture of both male and female DNA. These stains can be characterized by semen screening and DNA testing. Female DNA originates from epithelial cells from the vaginal wall, mouth or skin. Semen (male DNA) which contain up to 300 million sperm if deposited on bedsheets, clothing or undergarments can remain identifiable as dried stains for years. Material containing these stains can be quickly screened for presence semen or other fluid using three techniques: ultraviolet illumination, prostate specific antigen (p30) & microscopy. Once the stains are extracted and identified it is then compared to the male or female DNA reference.
